Bones combina humor e emoção mostrando o que se esconde por trás dos crimes mais aterrorizantes. No centro da trama está a Dra. Temperante Brennan (Emily Deschanel), uma excelente antropóloga forense que nas horas livres escreve romances. Ela é geralmente chamada pela policía quando o método padrão de identificação das vítimas não é suficiente. Sua habilidade para decifrar os mistérios ocultos nos corpos da vítima é única. Brennan geralmente trabalha com Seeley Booth (David Boreanaz), um agente da Unidade de Investigação de Homicídios do FBI. Booth não acredita na ciência e nos cientistas. Segundo ele, a chave para a solução dos crimes está em uma investigação à moda antiga, junto aos que ainda estão vivos, sejam eles suspeitos ou testemunhas. Com isso, Brennan e Booth frequentemente entram em choque, tanto no campo profissional como no âmbito pessoal.
